Job Description:

 

Perform laboratory procedures and experiments investigating the role of the airway remodeling and the extracellular matrix (ECM) in lung disease during acute and chronic airway inflammation using ex vivo primary human cell culture models. Technical skills include human primary airway epithelial cell, lung stromal cell, and immune cell tissue culture, differentiation of airway epithelial cells at an air-liquid interface, gene knockdown using lentiviral shRNA and/or gene editing using CRISPR/Cas9 methods in primary human cell lines, microscopy (phase contrast; fluorescent), biochemical assays, RNA isolation, qPCR, protein assays and ELISA.

 

Work includes infection of airway epithelial cells in a BSL-2 laboratory with respiratory viruses. Work as part of a small lab with close collaboration and support of a larger group of lab technicians, students, clinical research associates, and principal investigators.

 

Complete daily lab tasks including design and execution of experiments, maintenance of laboratory records, and opportunities for authorship on manuscripts.

Required Education/Experience:
- Bachelor’s Degree in an engineering or scientific discipline
- Prior experience in a laboratory setting; may include relevant educational programs.

Preferred:

 

At least one (1) year of experience in the following:
- Set up and maintenance of tissue culture; ideally primary cell culture
- RNA isolation, cDNA synthesis, quantitative PCR using TaqMan probes
- Conduct of protein and ELISA assays
- Use of shRNA or siRNA for gene knockdown experiments and/or CRISPR-Cas9 gene editing in primary cells
- Flow cytometry
- Experience with phase contrast and fluorescent microscopy

- Data entry and/or word processing experience

- Experience in analyzing data using spreadsheets and statistical analysis.
